In the Launch of the Screaming Narwhal, which part made you say "MONKEY ISLAND IS BACK!!"? You know, the defining moment that this is DEFINITELY Monkey Island.
For me, it's when we zoom out to the bird's eye view of Flotsam Island, classic MI style.:D

the opening credits and opening song. i actually had chills and goosebumps and a huge grin on my face the whole time
though i wish they had done it MI2 and CMI style with cool sepia drawings under the credits

This is what did it for me too, as soon as it zoomed out I grinned, because it's really back.

Yeah. The mix for the theme was neat(though not as neat as the one for LeChuck's Revenge), and the opening seemed to lack a bit of "flair" compared to the two games that seem to have had the most influence on Tales.

For me it was when you got the task of doing some newsworthy pirate stuff - starting a bar fight, finding a treasure and seizing the ship.
It all reminded me too well of the three trials. Awesome.

Well, all the way from the start it already felt like they managed to get it right to me, but what really convinced be beyond all doubt was the conversation with the Voodoo Lady: The options, the 'you sound familiar' bit at the door, more and more conversation options springing up so that you end up having a real 'good old' really long mostly pointless conversation with her. And of course grabbing some stuff from her hut and ruining stuff she was doing :).

The music is definitely the most iconic aspect of the MI theme for me. I still find myself humming it or the LeChuck music once in a while.
My first MI moment came when I got the first map puzzle. A nice variation on the classic "Do the Monkey" map in Secret.
Telltale did a great job of capturing the feel of MI, and I felt nostalgic the entire episode.

Also, the Chapter Screen did it for me. Probably the best think I've ever seen at one in the morning ;)

This is what did it for me also. For some reason, I wasn't expecting the island maps to be back. It's amazing how a little detail like that can make so much of a difference.

I was hoping to have some old tattered piratey illustrations blowing past in the foreground along with all of the other elements (this is why there are smaller pieces of paper in the background) but we just couldn't get it in along with everything else. Maybe for the Special Edition in 20 years :)
For me getting to put in the "Deep in the Caribbean" text is what made me realize that a MI game was going on here.

Yeah, that was pretty cool too. I just noticed how every game starts with those words. Though in this case, will it appear at the beginning of every episode?

Me too, when you're in front of the door of Voodoo Lady's place and the camera is put in a way that lets you see a close-up of a liana and some other vegetation (very cool!). Don't ask me why, but then I really said "OK, this is definitely pure Monkey Island."

Maybe in the second episode is "Deeper in the Caribbean", in the third "Even deeper in the Caribbean", and so on... :)

Yes it did.
The subtitles said it, but Guybrush said "From the personal log of Guybrush Threepwood"
Anyway, another thing that convinced me that this is Monkey Island is when I heard of La Esponja Grande (did I get it right?) these piratey legends are kinda trademark for MI (Big Whoop, Ultimate Insult)

for me it was Guybrush trying to make up an insult for sword fighting in the begining.
Even if he is cut by Elaine, I had a little moment of: "yeah!!!!"
