Not really. Almost none of the Telltale Games characters could work in a fighting game in any real meaningful way. Generally speaking, adventure game characters are adventure game characters because fighting isn't the(only) solution to their problems, and creating a moveset for them and making it feel natural is an impossible task.

Considering the only good telltale fighters would be Max, and Lechuck(if they ever get his rights) probably not.
Although someone, somewhere on the internetz said something about Nintendo vs Capcom.
That could work.

Who says it has to be a fighter? Namco X Capcom wasn't.

Still, the biggest problem is that Telltale doesn't actually have any characters.

Max and Sam could absolutely go crazy in a fighting game. Life low? Tag in Max for psychic power blasting and crazed biting action!

It could work, as a Super Smash Bros. clone. As a Street Fighter clone is would be a total fail, IMO.

Super Telltale Brothers Brawl Character Roster

Sam and Max:
*Sam- Similar to a Star fox Character, being able to kick and use his revolver. Final smash is the Desoto.
*Max- Cross between Kirby and a Star fox character. Final Smash is Maxthulu. (Don't worry, he'll get better.)
*Anton Papierwaite- Similar Moves to Zelda/Sheik. Final smash is a Rain of Dark Energy.
*Grandpa Stinky- Cross between Donkey kong and a Star Fox Character- Final Smash is similar to Kirby's cooking pot trick.

SBCG4AP:
*Strong Bad- Similar moves to Mario, which a touch of Captain Falcon. Final smash is a Heavy Metal Riff.
*Homestar Runner- Similar Moves to Sonic. Final smash is a Dangeresque-Style Spinning kick.
*Pom Pom- Similar to Kirby or King Dedede. His final smash is a flood of attractive women.
*Trogdor- Almost of clone of Bowser, with the kicking mechanism gone. His Final Smash is ULTRA-TROGDOR!!!
*Stinkoman- Basically a clone of Megaman, Borrowing some techniques from Samus. Final Smash is a giant lazor beam.
*1-Up- A clone of Homestar who borrows some moves from Stinkoman.-Final Smash is Similar to Homestar's.

Monkey Island:
*Guybrush Threepwood- Similar to Ike or Marth in terms of swordfighting. Final Smash is a game of Murray bowling.
*Gov. Elaine Marley- Similar to Zelda, with some Marth elements to her. Final smash is an offscreen pummeling
*Ghost/Zombie/Demon/??? Pirate LeChuck- Similar to Ganondorf. Final Smash is a Rain of pox.
*Reginald Van Winslow- Similar to Luigi, can also use his trusty map as a shield. Final smash is a cannon firing similar to Solid Snake.
*Morgan LeFlay- Similar to Elaine. Final Smash is a massive kick, similar to the movie 300.

Penny Arcade:
*Tycho- similar to Ness or Lucas from the Mother Series. Final smash is a 20-sided die roll.
*Gabe- similar to Tycho, with some minor altercations. Final smash is an FF 2000 which attacks the first enemy it can reach.

Team Fortress 2:
*Scout- Cross between Sonic and Ness Final smash is a blast from his "Force-a-Nature" shotgun.
*Heavy- Similar to Bowser, without the fire breathing, and can use shotguns. Final smash is Sasha.
*Spy- Can turn invisible and has knife wielding moves. Final Smash is a multiple back-stabbing move.
*Demoman- Similar to solid Snake. His final smash is similar to Ike's Great Ether.
*Soldier- similar to a Star Fox Character. his final smash is a bazooka launcher.

Puzzle Agent:
*Nelson Tethers- Similar to Mr. Game and Watch. His final smash is a crowd of hidden people.

Back to the Future:
*Marty McFly- Also similar to star fox, and can attack using his guitar. His final smash is the flying Delorean. (NOT for time travel.)
